Abstract

This study reports the incidence of histological alterations in morphometric changes in the liver and ovary of Acanthopagrus latus collected from an urban stream impaired by anthropogenic activities and from a clean site (reference). Mercury concentration was determined using a standard cold vapor atomic absorption. The results showed that mercury concentration at the creek waters, and specially sediments along Mahshahr coast, was higher than in the other marine environment, and in Zangi was lower than in the other sites of sampling. Several liver lesions were identified, including enlarged and lateral nuclei, nuclear degeneration and vacuolation, oncotic, apoptic, focal, massive, centrilobular and periportal  necrosis, atrophy, lipidosis, hydropic and cloudy swelling, oval cell proliferation, cirrhosis, hemorrhage, macrophage aggregates, bile stagnation, dilation of sinusoid, intracellular edema, and dark granules. Ovary histology revealed some changes in higher concentrations such as increase in the number of pre-vitellogenic oocytes, oocyte atresia, and adhesion in the ovaries of females that were sampled from the sites with high concentration of mercury. In this study, existence of mercury in Mahshahr coast is demonstrated and high incidence of histological alterations in the liver and ovary of A. latus is an evidence of the poor environmental quality of creek waters. Hence, histopathological changes were induced by the mercury adversely affected the proper functioning of these organs in these fish.

Abstract

The objective of this study was to evaluate the effect of probiotic, Pediococcus acidilactici (CNCM-MA 18/5 M, Lallemand, France) in concentrations of T1: 1 × 109, T2: 2× 109 and T3: 3×109  cfu / kg of diet on changes some hematological parameters of bream fry (Abramis brama orientalis, Berg 1949) in comparison with diet control, C, (untreated) for 60 days. One hundred eighty Bream fry with randomly in initial weight of 2.69± 0.22 g divided in 12 fiberglass tanks (110 L) and offered feed at 3-5% body weight, daily. At the end of the trial blood parameters measured. The total number of red blood cells, hematocrit, hemoglobin and white blood cells levels showed a significant increase in treatment fed with 2× 109  cfu probiotic per kilogram of diet (P<0.05).  Indifferential white blood cell(WBC) count, the number of neutrophils and monocytes, and lymphocytes increased significantly in treatments fed with probiotics. The highest increase in neutrophils and monocytes, and the highest decrease in lymphocytes observed in T3 (3× 109 cfu kg-1). According to the results, it concluded that adding probiotic Pediococcus acidilactici to the fry bream (Abramis brama orientalis, Berg 1949) diet, increase blood parameters related to red blood cells (RBC), diseases resistance and improve non specific immune response in bream.

Abstract

Oil pollution of waters, in addition to disrupting the physiological function of fish, leads to irreparable damage to human health during transmission through the food chain. Therefore, the possibility of accumulation of petroleum hydrocarbons in the edible tissues of fish is of particular importance. In this study, samples of yellowfin seabream (Acanthopagrus latus; Houttuyn, 1782) were collected from the Khowr-e Musa area in the Persian Gulf using bait traps. after twenty days of adaptation to the laboratory conditions (South of Iran Aquaculture Research Center), the fishes was divided into five groups with two replications in tanks containing 300 liters of seawater. The groups were then exposed to concentrations of 0, 2, 4, 8 and 16% water associated fraction (WSF).
The results of the study of the amount of total petroleum hydrocarbons in the gills and kidney tissue showed a significant increase on days 10 and 20 of the experiment, which increased slightly with increasing WSF concentration (P <0.05). While liver and muscle tissue, considering exposure time and WSF concentration, no significant difference was observed (P> 0.05). The results of the present study showed the possibility of accumulation of TPH in the gill and kidneys of yellowfin seabream, under the influence of concentration and duration of exposure to WSF. Therefore, considering the high economic value of the Sparidae family, it seems necessary to provide appropriate management methods to control oil pollution in the Persian Gulf.

Abstract

Piscidin has a wide range in killing microorganisms including bacteria, fungi, viruses, parasites and has strong anti-tumor activity and plays a role in increasing innate immunity and also does not provide resistance against bacteria; Therefore, it is of great importance in aquaculture. In this study, piscidin gene of Sparus aurata in vector pTZ57R / T was cloned. In this research, ligation product was transferred to component cell of E. coli DH5α strain. Plasmid extraction was performed from single colonies observed in ampicillin plate. Confirmation of the accuracy of single colonies grown in this research was performed by direct PCR and sequencing. The amplified cDNA fragment of the gilthead seabream piscidin gene consists of 310 nucleotides and 57 amino acids. The results of this research show that piscidin gene has been successfully cloned in pTZ57R / T vector. Comparison of nucleotide sequence of piscidin gene in this study showed high similarity with piscidin 5  of Morone chrysops. The comparison of the amino acid sequence of signal peptide piscidin is quite similar to Dicentracin-like of that species registered in the genebank, and mature peptide piscidin sequence is similar in only three amino acids to Pleorocidin-like of Poesila farmosa and Dicentracin-like of Sphaeramia orbicularis. This study could be a step towards further studies of piscidin peptide.

Abstract

The aim of this study was to investigate the effect of adding protein concentrate of Bream (Abramis Brama) on chemical properties, texture, color and sensory evaluation of pasta. The fillets were cooked in boiling water for 10 minutes, then minced using a meat grinder. The minced fish was dried at 60 C for 24 hours in an oven and powdered with a grinder. Pasta was prepared with different percentages of FPC (0, 5%, 7.5%, 10%, 15% and 20%). Chemical properties (energy, carbohydrate), tissue tests (hardness, adhesion and resilience), color tests (b, a, L) and sensory evaluation were evaluated. The results showed that by adding fish protein concentrate, the amount of pasta carbohydrate decreased compared to the control sample. In contrast, with increasing fish protein concentrate in pasta, the amount of energy increased significantly (p <0.05). The highest energy content was observed in the pasta containing 20% FPC and the lowest energy content in the control sample. The addition of FPC did not cause any adverse changes in the color of the pasta. The amount of yellow and redness of the pasta increased with the addition of FPC. The amount of pasta patency increased with the addition of fish protein concentrate. The color, taste, chewability, elasticity, odor, filament appearance, adhesion, hardness and overall evaluation of pasta with 7.5% FPC was better than others.  Regarding the texture, color and sensory characteristics of the pasta, it can be concluded that the pasta containing 7.5% fish protein concentrate had better characteristics than other samples.

Abstract

To aim health risk assessment and determination of consumption rate limit of King soldier bream (Argyrops spinifer), Pickhandle Barracuda (Sphyraena jello), and Japanese threadfin bream (Nemipteru japonicus), 33 fish specimen was harvest from Jofre fishing area. After transport fish to the laboratory, was prepared and concentration of heavy metal iron, manganese, and cobalt was measured by Inductively coupled plasma - optical emission spectrometry, (ICP-OES). The Concentration of heavy metal in the muscle of fish compared with international standards.  For health risk assessment Target Hazard Quotients (THQ) and Total Target Hazard Quotients (TTHQ), for determination of consumption rate limit estimated daily intake (EDI) and estimated weekly intake (EWI), Consumption Rate Limit (CRlim) and Consumption Rate Limit meal month (CRmm) in two groups include adults and children were calculated.  The Results showed that manganese content in all three fish species (36.59 to 55.48 µgr/gr) more than the world health organization (WHO) and the United States Environmental Protection Agency (USEPA). Also Cobalt content in Argyrops spinifer and Sphyraena jello (66.62 and 0.38 µgr/gr) more than WHO. According to THQ, TTHQ, EDI, and EWI, the Consumption of none of the three fish species had not health risk for the consumer. Additional to the highest CRlim in adults and children determinate as in Argyrops spinifer and Sphyraena jello was manganese, respectively (10.12 and 2.17 µgr/gr), (29.36 and 6.29 µgr/gr) and in Nemipteru japonicus (184.25 and 39.48 µgr/gr) Cobalt determinate.
